
Centrifugation medium made from polysucrose; density: 1.068 g/ml; for the isolation of human monocytesProduct descriptionOur BioColl® separation solution contains neutral, highly cross-linked polysucrose with an average molecular weight of 400,000 Da. This hydrophilic polymer can be used to prepare aqueous solutions with different densities (up to about 1.2 g/ml), which are used for the isolation of cells or subcellular particles. The appropriate density of the BioColl® separation solution is selected depending on the particle type. A density of 1.068 g/ml is ideal for the isolation of human monocytes. For optimal pH and osmolality values, the BioColl® separation solution should be adjusted with an acid (preferably amidotrizoic acid (ATS) ) and sodium hydroxide solution.In order to specifically separate monocytes from other mononuclear blood cells (such as lymphocytes), a double polysucrose gradient is required. For this purpose, BioColl® for monocytes (BS.L6815) is layered over BioColl® for lymphocytes (BS.L6115) . Due to their higher density, lymphocytes and thrombocytes pass through the BioColl® layer for monocytes (density 1.068 g/ml), but not the BioColl® layer for lymphocytes (density 1.077 g/ml) . After centrifugation, they are located between the two BioColl® layers. Monocytes do not penetrate the BioColl® layer for monocytes (density 1.068 g/ml) due to their lower density. After centrifugation, they are located between the BioColl® for monocytes (density 1.068 g/ml) and the plasma layer.
